Latest genera- tions of microscopes, able to produce huge arrays of multidimensional data, only distantly resemble Leeuwenhoek’s first microscope. Every advance in visualization techniques and hardware brings us one step closer to understanding life, e.g., how genome information gives identity to cells, how cells constitute organisms and how errant cells cause disease. Discovery of the green fluorescent protein (GFP) in the nineties of the previous century was definitely one of the most impor- tant milestones on that path, giving new strong impulse to the field of fluorescence microscopy

    Joint Intensity Inhomogeneity Correction for Whole-Body MR Data

    Abstract. Whole-body MR receives increasing interest as potential alternative to many conventional diagnostic methods. Typical whole-body MR scans contain multiple data channels and are acquired in a multistation manner. Quantification of such data typically requires correction of two types of artefacts: different intensity scaling on each acquired image stack, and intensity inhomogeneity (bias) within each stack. In this work, we present an all-in-one method that is able to correct for both mentioned types of acquisition artefacts. The most important properties of our method are: 1) All the processing is performed jointly on all available data channels, which is necessary for preserving the relation between them, and 2) It allows easy incorporation of additional knowledge for estimation of the bias field. Performed validation on two types of whole-body MR data confirmed superior performance of our approach in comparison with state-of-the-art bias removal methods

    The effect of mirabegron on energy expenditure and brown adipose tissue in healthy lean South Asian and Europid men

    Aim: To compare the effects of cold exposure and the β3-adrenergic receptor agonist mirabegron on plasma lipids, energy expenditure and brown adipose tissue (BAT) activity in South Asians versus Europids. Materials and Methods: Ten lean Dutch South Asian (aged 18-30 years; body mass index [BMI] 18-25 kg/m2 ) and 10 age- and BMI-matched Europid men participated in a randomized, double-blinded, cross-over study consisting of three interventions: short-term (~ 2 hours) cold exposure, mirabegron (200 mg one dose p.o.) and placebo. Before and after each intervention, we performed lipidomic analysis in serum, assessed resting energy expenditure (REE) and skin temperature, and measured BAT fat fraction by magnetic resonance imaging. Results: In both ethnicities, cold exposure increased the levels of several serum lipid species, whereas mirabegron only increased free fatty acids. Cold exposure increased lipid oxidation in both ethnicities, while mirabegron increased lipid oxidation in Europids only. Cold exposure and mirabegron enhanced supraclavicular skin temperature in both ethnicities. Cold exposure decreased BAT fat fraction in both ethnicities. After the combination of data from both ethnicities, mirabegron decreased BAT fat fraction compared with placebo. Conclusions: In South Asians and Europids, cold exposure and mirabegron induced beneficial metabolic effects.
